目的 探讨玻璃体腔注射乳酸环丙沙星治疗方法对视网膜组织的安全性。 方法 2 4只兔随机分为4组,每组6只。玻璃体腔内分别注入0 .1ml不同浓度的乳酸环丙沙星,浓度分别为:2 5 0 0、5 0 0 0、10 0 0 0 μg/ml;对照组注射生理盐水0 .1ml。手术后观察眼底表现,分别行光学显微镜、透射电子显微镜等组织学检查和视网膜电图(ERG)视网膜功能检查。 结果 2 5 0、5 0 0 μg组光学显微镜检查结果未见异常,超微结构大致正常;10 0 0 μg组光学显微镜下可见视网膜内外核层排列不规则,神经节细胞水肿变性或脱失,透射电子显微镜下可见超微结构改变。视网膜功能检查显示,各组ERG的a~b波波幅注药前后无明显差别。 结论 玻璃体腔注射国产乳酸环丙沙星安全、抗菌谱广,5 0 0 μg以下是较为安全的眼内应用剂量。
Objective To investigate the safety of intravitreal injection of ciprofloxacin lactate in retinal tissue. Methods Twenty-four rabbits were randomly divided into 4 groups with 6 rats in each group. Into the vitreous cavity were injected with 0.1ml different concentrations of ciprofloxacin lactate, the concentrations were: 255050000, 101000μg / ml; the control group injected with normal saline 0.1ml. The ocular fundus was observed after operation. The histopathological examination such as optical microscope and transmission electron microscopy, and the retinal electroretinogram (ERG) were performed respectively. Results There was no abnormality in the microscopic examination of 250, 500 microg / kg microgroups, and the ultrastructure was almost normal. Under 10 000 microgroups, the inner and outer nuclear layers of the retina were arranged irregularly, the degeneration or loss of ganglion cell edema, Transmission electron microscopy shows ultrastructural changes. Retinal function tests showed that there was no significant difference in a ~ b wave amplitude of ERG between groups before and after drug injection. Conclusion The intravitreal injection of homemade ciprofloxacin lactate is safe and broad in antibacterial spectrum. The dosage below 500 μg is safe for intraocular application.
