目的建立稳定、成熟的内皮细胞原位获取法。方法应用原位获取法从恒河猴及成人的73条表浅或腹腔血管中获取内皮细胞。A、B两组分别用0.1%Ⅱ型胶原酶消化25分钟、15分钟。结果A组内皮细胞获取率明显高于B组。两组中大隐静脉、劲外静脉、上肢浅静脉、胃网膜右静脉、髂总动脉的内皮细胞获取率明显高于既往的机械获取法、灌注酶解消化法、外翻酶解消化法(P<0.01).结论原位获取法的内皮细胞获取率明显高于既往的内皮细胞获取方法,其理想的消化时间25分钟。
Objective To establish a stable and mature endothelial cells in situ method. Methods Endothelial cells were obtained from 73 superficial or peritoneal blood vessels of rhesus macaques and adults using in situ acquisition. Groups A and B were digested with 0.1% collagenase for 25 minutes and 15 minutes, respectively. Results A group of endothelial cell acquisition rate was significantly higher than the B group. In both groups, the acquisition rate of endothelial cells in great saphenous vein, extragranular vein, superficial vein of upper limb, right gastric vein and common iliac artery was significantly higher than those of mechanical acquisition, perfusion digestion, (P0.01) .Conclusion The acquisition rate of endothelial cells by in situ method is obviously higher than that of previous methods of obtaining endothelial cells, and the ideal digestion time is 25 minutes.
