[目的]探讨沙利度胺对人鼻咽癌细胞株细胞周期分布及体外放射敏感性的影响。[方法]应用MTT法检测不同时间点、不同浓度沙利度胺对人鼻咽癌细胞株CNE-2细胞增殖的抑制率。采用沙利度胺及放射线单独或联合作用于CNE-2细胞,用克隆形成法经单击多靶模型拟合细胞存活曲线,获得不同处理条件下的放射增敏比;流式细胞技术分析沙利度胺对CNE-2细胞周期分布的影响。[结果]沙利度胺可抑制CNE-2细胞的增殖,且呈浓度依赖性;20mg/L和40mg/L沙利度胺作用CNE-2细胞24h后均见到放射增敏作用,SER分别为1.29(0.941/0.728)和1.57(0.941/0.598);流式细胞仪分析表明沙利度胺作用24h,随着沙利度胺浓度的增加,CNE-2细胞中处于G0/G1期的细胞增多,S期和G2/M期细胞减少,但变化差异无显著性。[结论]沙利度胺能够抑制CNE-2细胞的增殖,并具有一定的放射增敏作用,为临床放疗和沙利度胺的联合应用提供了实验依据。
[Objective] To investigate the effect of thalidomide on cell cycle distribution and radiosensitivity in human nasopharyngeal carcinoma cell lines. [Method] MTT assay was used to detect the inhibitory rate of thalidomide on the proliferation of human nasopharyngeal carcinoma cell line CNE-2 at different time points. CNE-2 cells were treated with thalidomide and radiation alone or in combination, and clonal formation method was used to fit the cell survival curve by single-click multi-target model to obtain the radiosensitization ratio under different treatment conditions; flow cytometry analysis of sand Effect of Lidosis on the Cell Cycle Distribution of CNE-2 Cells. [Result] Thalidomide could inhibit the proliferation of CNE-2 cells in a concentration-dependent manner. Radiosensitization was observed in CNE-2 cells treated with 20 and 40 mg / L thalidomide for 24 h, respectively (1.94 (0.941 / 0.728)) and 1.57 (0.941 / 0.598), respectively. Flow cytometry analysis showed that thalidomide increased the concentration of thalidomide and the cells in G0 / G1 phase of CNE-2 cells Increased, S phase and G2 / M phase cells decreased, but the difference was not significant. [Conclusion] Thalidomide can inhibit the proliferation of CNE-2 cells and has some radiosensitization effects, which provides experimental evidence for the combination of clinical radiotherapy and thalidomide.
