目的研究孕鼠妊娠晚期应用不同疗程地塞米松治疗对大鼠子代脑神经元凋亡的影响。方法 SPF级SD孕大鼠30只,随机分为3组,产前1剂组10只,于孕17 d肌注地塞米松0.8 mg/(kg·d);产前4剂组10只,于孕17、18、19、20 d各肌注地塞米松0.8 mg/(kg·d);对照组10只肌注等量生理盐水。对各组孕鼠所生仔鼠,分别于生后第1、7、14 d每组每个时间点随机取10只进行脑神经元凋亡细胞的原位标记检测,并与对照组比较。结果产前4剂组生后1、7 d仔鼠与对照组相比,神经元凋亡增加,差异有极显著性(P<0.01);产前1剂组与对照组相比无明显差别 (P>0.05)。结论重复产前激素治疗(ACT)会引起仔鼠脑神经元凋亡增加,影响仔鼠脑发育。
Objective To study the effects of dexamethasone on the apoptosis of rat neurons in the third trimester of pregnancy using different courses of dexamethasone. Methods Thirty SPF-SD pregnant rats were randomly divided into 3 groups, 10 pre-natal group and 10 dexamethasone 0.8 mg / (kg · d) Only intramuscular injection of dexamethasone 0.8 mg / (kg · d) on the 17th, 18th, 19th and 20th day of gestation. The control group was injected intraperitoneally with the same amount of normal saline. Results Compared with the control group, the apoptosis rate of neurons in the 4 groups of prenatal 4 groups was significantly higher than that of the control group (P <0.01). Conclusions Repeated prenatal hormonal therapy (ACT) causes increased neuronal apoptosis in the brains of the offspring and affects brain development in the offspring.
