【目的】通过比较血塞通注射液等在类过敏试验中致RBL-2H3细胞与大鼠腹腔原代肥大细胞脱颗粒特性的异同,以评价2种细胞在中药注射液类过敏试验中的意义。【方法】选用已经明确能引起类过敏反应的3种供试品即血塞通注射液、Compound 48/80和吐温80,分别与RBL-2H3细胞和大鼠腹腔肥大细胞共同孵育1 h,通过β-氨基己糖苷酶释放率和组胺释放率、释放量等的检测,分析比较脱颗粒特性的异同,同时观察了不同浓度血塞通注射液对脱颗粒程度的影响。【结果】2种细胞在血塞通注射液、Compound 48/80和吐温80的直接刺激下均出现脱颗粒现象,脱颗粒程度与血塞通注射液呈剂量依赖性;原代肥大细胞β-氨基己糖苷酶释放率、组胺释放量和释放率均显著高于RBL-2H3细胞,2种细胞比较差异有统计学意义(P<0.01)。【结论】在类过敏反应脱颗粒试验中,RBL-2H3细胞和原代大鼠腹腔肥大细胞脱颗粒具有相同趋势,均可用于中药注射液类过敏评价,RBL-2H3细胞应用方便,而肥大细胞更加敏感。
【Objective】 The purpose of this study is to compare the desensitization of primary mast cells between RBL-2H3 cells and rat peritoneal cavity in the class of anaphylactic allergy test such as Xuesaitong injection to evaluate the significance of the two kinds of cells in the allergy test of traditional Chinese medicine injection . 【Method】 Three kinds of test materials, ie, Xuesaitong injection, Compound 48/80 and Tween 80, which were already able to cause allergic reaction, were selected and incubated with RBL-2H3 cells and rat peritoneal mast cells for 1 h, Through the detection of β-hexosaminidase release rate, histamine release rate and release amount, the similarities and differences of degranulation characteristics were analyzed and compared. Meanwhile, the effect of different concentrations of Xuesaitong injection on degranulation degree was observed. 【Results】 The two kinds of cells showed degranulation under the direct stimulation of Xuesaitong injection, Compound 48/80 and Tween 80. The degree of degranulation was in a dose-dependent manner with Xuesaitong injection. The proliferation of primary mast cells β - hexosaminidase release rate, histamine release and release rate were significantly higher than RBL-2H3 cells, the two kinds of cells was statistically significant difference (P <0.01). 【Conclusion】 In the anaphylactoid degranulation test, the degranulation of peritoneal mast cells in RBL-2H3 cells and primary rat have the same trend, which can be used to evaluate the allergic reaction of Chinese medicine injection. The application of RBL-2H3 cells is convenient, while the mast cells More sensitive.
