目的建立测定猫豆中左旋多巴及其衍生物1-甲基-3-羧基-6,7-二羟基-1,2,3,4-四氢异喹啉(MCD)含量的亲水作用高效液相色谱法。方法采用Cosmosil HILIC色谱柱(4.6 mm×250 mm,5μm),柱温30℃,以乙腈-(10 mmol/L)乙酸铵(80∶20)为流动相,体积流量1.0 ml/min,检测波长280 nm。结果 MCD及左旋多巴在0.081 36~2.034μg和0.126 9~3.172μg范围内线性关系良好,平均回收率(n=6)分别为98.11%和99.86%,相对标准偏差(RSD)分别为3.32%和0.82%。结论该方法简便、准确,可用于定量分析猫豆中的左旋多巴及MCD。
Objective To establish a hydrophilicity assay for the determination of levodopa and its derivatives 1-methyl-3-carboxy-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline (MCD) High performance liquid chromatography. Methods Cosmosil HILIC column (4.6 mm × 250 mm, 5 μm) was used with a mobile phase of acetonitrile (10 mmol / L) ammonium acetate (80:20) at a flow rate of 1.0 ml / 280 nm. Results MCD and levodopa showed good linearity in the range of 0.081 36-2.034μg and 0.126 9-3.17-2μg with the average recoveries (n = 6) of 98.11% and 99.86%, respectively, and the relative standard deviations (RSDs) were 3.32% And 0.82%. Conclusion The method is simple and accurate and can be used for quantitative analysis of levodopa and MCD in cat bean.
