目的 探讨利用细胞涂片行P—糖蛋白(P—glycoprotein,Pgp)检测的可行性。方法 用肿瘤病人的胸腹水,穿刺涂片等细胞学标本,以链霉菌抗生物素蛋白—碱性磷酸酶(Streptavidin—alkaline phosphatase,SAP)免疫细胞化学方法检测肿瘤细胞的Pgp表达。结果 各种不同涂片标本肿瘤细胞的Pgp表达总阳性率为31.9%,阳性表达定位准确,呈色鲜艳清晰,对比鲜明。结论 用SAP法检测各种细胞学标本的Pgp表达,可为不能手术的,晚期的以及使用耐药逆转剂的恶性肿瘤患者提供测定其耐药状态的简便、实用的方法。
Objective To investigate the feasibility of P-glycoprotein (Pgp) detection by using cell smears. Methods Pgp expression in tumor cells was detected by streptoavidin-alkaline phosphatase (SAP) immunocytochemistry using cytology specimens from thoracentesal fluid and puncture smears of tumor patients. Results The positive rate of Pgp expression in tumor cells was 31.9% in all smear specimens. The positive expression was accurate, the color was bright and clear, and the contrast was clear. Conclusion The detection of Pgp expression in various cytological specimens using the SAP method can provide a simple and practical method for determining the drug resistance of malignant tumor patients who are inoperable, advanced, and who have used drug resistance reversal agents.
