黄花夹竹桃果仁用石油醚脫脂后,采用发酵法提取总单糖甙。总单糖甙經氧化鋁柱层离,除分得neriifolin和peruvoside以外,又分出另三个强心甙A,D,E,强心甙A証明是cerberin,C_(32)H_(48)O_9,熔点为215—218℃,[α]_D~(29)—82.4(c=1.1412,甲醇),U.V.λ_(max)218毫微米(logε,4.19),IR(KBr)——2.96,5.57,5.67—5.78,6.20,7.25,7.82—8.0,9.80微米。强心甙D証明是ruvoside,C_(30)H_(46)O_9,熔点为239—240%,[α]_D~(29)—55.5(c=1.2727,甲醇),U.V.λ_(max)218毫微米(logε,4.21),IR(KBr)—2.97,5.79,6.20,7.25,9.80微米。强心甙E,C_(30)H_(44)O_(10),熔点为168—170℃,[α]_D~(28.5)—45.47(c=1.37,甲醇),pK_α 6.2,U.V.λ_(max)218毫微米(logε 4.23),IR(KBr)——2.98,5.59,5.72,5.90,6.16,7.26,9.75,微米,其三乙胺盐的紅外光譜出現6.45微米的吸收峯而5.90微米的吸收峯消失。peruvoside 在丙酮中用高錳酸鉀氧化后所得产物的物理化学性貭与强心甙E完全一致,故証明强心甙E为一个新强心甙,其結构式为:cannogeninic acid—l—thevetoside,并命名为perusitin。
Yellow oleander nectar kernels were degreased with petroleum ether and total monosaccharide glycosides were extracted by fermentation. Total monosaccharide glycosides were separated by alumina column. In addition to the nerifolin and peruvoside, three other cardiac glycosides A, D, and E were isolated. The cardiac glycosides proved to be cerberin, C_(32)H_(48). O_9, melting point 215-218°C, [α]_D 29-82 (c=1.1412, methanol), UV λ max (max 218 nm (log ε, 4.19), IR (KBr) - 2.96, 5.57 5.67-5.78, 6.20, 7.25, 7.82-8.0, 9.80 microns. The strong heart 甙D proved to be ruvoside, C_(30)H_(46)O_9, the melting point was 239—240%, and [α]_D~(29)—55.5 (c=1.2727, methanol), UVλ_(max) 218 mM. Micron (logε, 4.21), IR (KBr)—2.97, 5.79, 6.20, 7.25, 9.80 μm. Strong cardiac enthalpy E, C_(30)H_(44)O_(10), melting point 168-170°C, [α]_D~(28.5)—45.47(c=1.37, methanol), pK_α 6.2, UVλ_(max ) 218 nm (log ε 4.23), IR (KBr) - 2.98, 5.59, 5.72, 5.90, 6.16, 7.26, 9.75, micron, the infrared spectrum of its triethylamine salt showed an absorption peak of 6.45 μm and absorption of 5.90 μm. The peak disappeared. The physicochemical properties of peruvoside obtained by oxidation with potassium permanganate in acetone are completely consistent with that of strong cardiac enthalpy E. Therefore, it is proved that the strong cardiac glycoside E is a new cardiac glycoside and its structural formula is: cannogeninic acid—l—thevetoside And named perusitin.
