目的探讨灵芝孢子粉对荷Lewis肺癌瘤株小鼠突变型P53基因表达的影响及抑瘤作用。方法将荷Lewis肺癌瘤株小鼠分别用100%和50%浓度剂量的灵芝孢子粉0.4 mL灌胃14 d。停药后处死小鼠,检测瘤重,计算灵芝孢子粉对荷Lewis肺癌的抑瘤率,并采用标记的链白素-生物素辣根过氧化酶复合物法检测瘤组织突变型P53蛋白,结果服用灵芝孢子粉的肺癌小鼠,抑瘤率分别为41.6%和39.9(P<0.01);突变型P53蛋白表达阳性细胞显著降低(P<0.01)。结论灵芝孢子粉能抑制肺癌小鼠癌基因P53的表达,并有较强的抑瘤作用。
Objective To investigate the effect of Ganoderma lucidum spore powder on the expression of mutant P53 gene in mice bearing Lewis lung carcinoma and its anti-tumor effect. Methods Mice bearing Lewis lung cancer were inoculated intragastrically with 0.4 mL ganoderma lucidum spores powder of 100% and 50% respectively for 14 days. The mice were sacrificed and the tumor weight was measured. The inhibition rate of Ganoderma lucidum spore powder on Lewis lung carcinoma was calculated. The mutant P53 protein was detected by the labeled streptavidin-biotin-horseradish peroxidase complex method. Results The inhibitory rate of lung cancer in mice treated with Ganoderma lucidum spores was 41.6% and 39.9, respectively (P <0.01). The expression of mutant P53 protein was significantly decreased (P <0.01). Conclusion Ganoderma lucidum spore powder can inhibit the expression of oncogene P53 in lung cancer mice and has strong anti-tumor effect.
