多巴胺是一种在机体内起重要调节作用的生物活性胺。为了对组织中的多巴胺进行免疫组织化学定位和放射免疫定量研究,作者用多巴胺-牛血清白蛋白复合物作为免疫原,免疫新西兰大白兔,成功地制备了抗多巴胺抗血清。用ABC免疫组织化学法,在经Bouin液固定的豚鼠消化道常规石蜡切片上检测,抗血清的效价为1∶1000～1∶2000。替代试验、吸收试验和交叉吸收试验证明,抗血清所显示的阳性染色为多巴胺抗体与组织中多巴胺的特异性免疫反应。
Dopamine is a bioactive amine that plays an important regulatory role in the body. In order to quantitatively study the immunohistochemistry and radioimmunoassay of dopamine in tissues, the authors immunized New Zealand white rabbits with dopamine-bovine serum albumin complex as immunogen and successfully prepared anti-dopamine antiserum. Using ABC immunohistochemistry, the antiserum titer was determined to be 1: 1000 to 1: 2000 on bouin-fixed guinea pig gastrointestinal routine paraffin sections. Substitution tests, absorption tests and cross-absorption tests demonstrated that the positive staining exhibited by the antiserum was a specific immune response of dopamine to dopamine in the tissues.
