目的探讨维生素E(Ve)及叶酸对高血糖诱导的乳鼠心肌细胞凋亡蛋白caspase-3表达的影响。方法将50只清洁级成年SD孕鼠随机分为5组,分别为正常对照组(A组)、妊娠糖尿病组(B组)、叶酸组(C组)、维生素E组(D组)和Ve+叶酸组(E组)。除对照组外,其余大鼠于妊娠d 1腹腔注射链脲佐菌素(STZ)35 mg/kg制备妊娠糖尿病动物模型,对照组给予等量0.1 mmol/L(p H 4.5)柠檬酸钠—柠檬酸缓冲液。孕d 5,D、E 2组予50 mg/kg维生素E灌胃,C、E组予0.4 mg/kg叶酸灌胃,A、B组予等量的生理盐水灌胃,持续7 d。取自然分娩的乳鼠心肌组织于光镜下观察心肌结构的改变,应用免疫组织化学法检测各组乳鼠心肌细胞caspase-3抗原的表达。蛋白免疫印迹(western-blot)检测各组乳鼠心肌细胞caspase-3的表达。结果 (1)造模前各组血糖值比较差异无统计学意义(P>0.05);造模后,B、C、D、E组血糖值明显高于A组,差异有统计学意义(P<0.01);B、C、D、E 4组间比较差异无统计学意义(P>0.05)。(2)光镜下观察B、C、D、E组乳鼠心肌组织呈现不同程度损伤,表现为细胞坏死、水肿、轮廓不清,心肌纤维排列疏松紊乱、炎性细胞浸润。(3)5组乳鼠心肌细胞凋亡蛋白caspase-3的表达不同,差异有统计学意义(P<0.01)。结论妊娠糖尿病孕鼠对子代心肌细胞凋亡影响明显,孕期补充维生素E、叶酸对乳鼠心肌细胞凋亡有明显改善作用,且叶酸抑制凋亡的作用优于维生素E,同时补充维生素E和叶酸的效果优于单独补充一种。
Objective To investigate the effects of vitamin E and folic acid on the expression of caspase-3 in neonatal rat cardiomyocytes induced by hyperglycemia. Methods Fifty clean adult pregnant SD rats were randomly divided into five groups: normal control group (group A), gestational diabetes mellitus group (group B), folic acid group (group C), vitamin E group (group D) and Ve + Folic acid group (E group). Except the control group, the other rats were intraperitoneally injected with streptozotocin (STZ) at a dose of 35 mg / kg for gestational diabetes mellitus, and the control group was given 0.1 mmol / L (p H 4.5) sodium citrate - Citric acid buffer. Pregnant d 5, D, E 2 groups were given 50 mg / kg vitamin E orally, group C, E were given 0.4 mg / kg folic acid, group A, B were given the same amount of saline for 7 d. Myocardial tissue of spontaneous childbirth was observed under light microscope, and the changes of myocardial structure were observed. The expression of caspase-3 antigen in cardiomyocytes was detected by immunohistochemical method. Western-blot was used to detect the expression of caspase-3 in neonatal rat cardiomyocytes. Results (1) There was no significant difference in blood glucose between the two groups before modeling (P> 0.05). After modeling, the blood glucose of group B, C, D and E were significantly higher than that of group A (P <0.01). There was no significant difference between B, C, D and E 4 groups (P> 0.05). (2) Under light microscope, the myocardial tissues in groups B, C, D and E showed different degrees of injury, which showed necrosis, edema, unclear outline, loosely arranged myocardial fibers and infiltration of inflammatory cells. (3) The expression of caspase-3 in 5 groups of neonatal rat cardiomyocytes was different, the difference was statistically significant (P <0.01). Conclusion Pregnant gestational diabetes mellitus has a significant effect on the apoptosis of myocardial cells of offspring. Vitamin E supplementation and folic acid during pregnancy significantly improve the apoptosis of neonatal rat cardiomyocytes, and the effect of folic acid on apoptosis is better than that of vitamin E, while vitamin E and Folic acid is better than a single supplement.
