目的 探讨细胞周期调控因子P16 -cyclin -Rb的通路与膀胱癌关系。 方法 应用免疫组织化学方法检测P16、Rb和cyclinD1在膀胱癌和正常膀胱组织中表达。 结果 P16和Rb在膀胱癌中表达显著低于正常组织 ,而且与肿瘤分级、分期密切相关 ,但与肿瘤复发无显著相关性 ;cyclinD1在膀胱癌中表达显著高于正常组织 ,但与肿瘤分级、分期和复发之间无显著相关性。膀胱癌中三种蛋白总异常率为 97.2 % ,其中同时两种或三种蛋白异常率达 6 1.1% ,肿瘤复发率也增高。膀胱癌中P16与Rb和cyclinD1分别呈负相关表达。结论 P16 -cyclin -Rb的通路异常与膀胱癌的发病机制密切相关 ,同时检测三者有助于更好了解膀胱癌生物学行动。
Objective To investigate the relationship between the cell cycle regulatory factor P16-cyclin-Rb pathway and bladder cancer. Methods Immunohistochemistry was used to detect the expression of P16, Rb and cyclinD1 in bladder cancer and normal bladder tissues. Results The expression of P16 and Rb in bladder cancer was significantly lower than that in normal tissues, and was closely related to tumor grade and stage, but not to tumor recurrence. The expression of cyclinD1 in bladder cancer was significantly higher than that in normal tissues, There was no significant correlation between staging and recurrence. The total abnormal rate of three proteins in bladder cancer was 97.2%, of which two or three proteins simultaneously anomalous rate of 61.1%, tumor recurrence rate also increased. There was a negative correlation between P16, Rb and cyclinD1 in bladder cancer. Conclusions The abnormal P16-Cyclin-Rb pathway is closely related to the pathogenesis of bladder cancer. The simultaneous detection of these three genes may help to understand the biological behavior of bladder cancer.
