健脾补肾药对脾虚大鼠环核苷酸及甲状腺激素水平的影响(英文)

背景:健脾补肾中药对实验性脾虚证有较好的防治作用,环核苷酸及甲状腺激素水平的变化是否与药物作用机制有关?目的:观察健脾补肾方药对实验性脾虚证大鼠环核苷酸及甲状腺激素水平的影响,探讨该方防治脾虚证的作用机制及脾与肾相互关系的意义。设计:随机对照的实验研究。地点和材料:实验地点为广州中医药大学核医学教研室。实验动物为雄性SD大鼠,体质量190~230g,3月龄,普通级,由中山大学实验动物中心提供(动物合格证号为2001A032)。干预:将大鼠随机分为正常对照组(n=10)、脾虚模型组(n=12),健脾补肾方高剂量组(n=12)和健脾补肾方低剂量组(n=12),采用大黄复制脾虚证动物模型,分别给予生理盐水、大黄、健脾补肾方煎液(高剂量:22g/kg,低剂量:11g/kg)灌胃。各组动物于最后1次给药后1h从眼球取血,观察各组大鼠血浆环磷酸腺苷(cyclicadenosinemonophos-phate,cAMP),环磷酸鸟苷(cyclicguanosinemonophosphate,cGMP),及甲状腺素(thyroxine,T4)、三碘甲腺原氨酸(thiiodothyronine,T3)、促甲状腺素(thyroidstimulatinghormone,TSH)水平的变化。主要观察指标:各组大鼠cAMP,cGMP,T4,T3,TSH水平。结果:脾虚模型组大鼠血浆cAMP含量犤(30.29±6.55)nmol/L犦及cAMP/cGMP(2.94±1.13)比值均比正常对照组升高,T3犤(1.55±0 BACKGROUND: The traditional Chinese medicine for strengthening the spleen and tonifying the kidney has good preventive and therapeutic effects on experimental spleen deficiency syndrome, and whether the changes of cyclic nucleotides and thyroid hormone levels are related to the mechanism of drug action. Design: Randomized controlled experimental study. Location and Materials: The experimental site is the Department of Nuclear Medicine, Guangzhou University of Chinese Medicine. Experimental animals were male SD rats, body weight 190-230 g, 3 months old, general grade, provided by the Laboratory Animal Center of Sun Yat-sen University (animal certificate number 2001A032). Intervention: Rats were randomly divided into normal control group (n=10), spleen deficiency model group (n=12), high-dose Jianpi Bushenfang group (n=12) and low-dose group of Jianpi Bushenfang (n=12). Rhubarb was used to replicate the spleen-deficiency syndrome animal model and was given intragastrically with normal saline, rhubarb, Jianpi Bushen decoction (high dose: 22g/kg, low dose: 11g/kg). Blood samples were taken from the eyeball 1 hour after the last administration in each group of animals. Cyclic adenosine monophos-phate (cAMP), cyclic guanosine monophosphate (cGMP), and thyroxine were observed in each group of rats. T4) Changes in the levels of thiiodothyronine (T3) and thyroidstimulating hormone (TSH). MAIN OUTCOME MEASURES: Levels of cAMP, cGMP, T4, T3, and TSH in each group of rats.
