[目的]研究白藜芦醇(Resveratrol,Res)对食管癌Eca109细胞的抑制作用。[方法]将食管癌Eca109细胞分为4组:Res1组、Res2组、Res3组和对照组,Res浓度分别为10、20、40和0μmol/L。用MTT法测定Res对Eca109细胞的抑制作用,采用RT-PCR方法检测Eca109细胞Caspase-3、Fas基因表达情况。[结果]10μmol/LRes处理24h对Eca109细胞有明显的抑制作用;20μmol/LRes处理48h后抑制作用达到极显著水平。20μmol/L和40μmol/LRes处理72h,Eca109细胞Caspase-3、Fas基因表达明显高于对照。[结论]Res可显著抑制Eca109细胞增殖,Res可通过促进食管癌细胞株中Caspase-3、Fas的表达发挥其抗肿瘤作用。
[Objective] To investigate the inhibitory effect of resveratrol (Resveratrol, Res) on esophageal carcinoma Eca109 cells. [Methods] The esophageal cancer Eca109 cells were divided into 4 groups: Res1 group, Res2 group, Res3 group and control group. Res concentrations were 10, 20, 40 and 0 μmol / L, respectively. The inhibitory effect of Res on Eca109 cells was determined by MTT method. The expression of Caspase-3 and Fas genes in Eca109 cells was detected by RT-PCR. [Result] The treatment with 10μmol / LRes for 24 h had a significant inhibitory effect on Eca109 cells, and the inhibitory effect reached a significant level at 48 h after treatment with 20 μmol / L res. The mRNA expression of Caspase-3 and Fas in Eca109 cells was significantly higher than that in control at 20μmol / L and 40μmol / LRes for 72h. [Conclusion] Res can significantly inhibit the proliferation of Eca109 cells. Res can play an antitumor role by promoting the expression of Caspase-3 and Fas in esophageal cancer cell lines.
