目的观察水通道蛋白7(aquaporin-7,AQP-7)在小鼠肾小管发育过程中的表达,探讨其与肾小管发生发育的关系。方法选取胚龄(E)12、14、17、18 d胎鼠及生后(P)1、3、7、14、24、40、70 d仔鼠肾脏,应用免疫组织化学技术观察AQP-7在肾小管中的表达;体视学方法测量AQP-7阳性表达的面密度值;应用免疫印迹技术检测AQP-7在各发育阶段肾组织中的含量变化。结果免疫组化结果显示AQP-7在E14 d表达于发育中的肾小管,随后表达在近端小管,P14 d后主要定位在皮质和外髓部近直小管的刷状缘,生肾区与內髓则无阳性表达;体视学测量结果显示随着胚(日)龄的增加,AQP-7在肾小管管腔面的面密度值逐渐增加,P24 d达最大值,后趋于稳定;免疫印迹结果证实,AQP-7在肾的表达量逐渐增加,P24 d达峰值后趋于稳定。结论 AQP-7在小鼠肾小管发育过程中存在时空性的表达,对发育后期小鼠肾的水和甘油平衡起重要调节作用。
Objective To observe the expression of aquaporin-7 (AQP-7) during the development of renal tubules in mice and to explore its relationship with the development of renal tubules. Methods The fetuses of embryonic age (E) 12, 14, 17 and 18 days and postnatal days 1, 3, 7, 14, 24, 40 and 70 were used to observe the effects of AQP-7 The expression of AQP-7 in renal tubules was detected by stereological methods. The changes of AQP-7 in renal tissues at various developmental stages were detected by Western blotting. Results The results of immunohistochemistry showed that AQP-7 was expressed in the developing tubules on E14 d and then expressed in the proximal tubules. After P14 d, AQP-7 mainly located in the brush border of cortical and extramedullary near straight tubules, The results of stereological measurement showed that the surface density of AQP-7 in the renal tubular surface increased gradually with the increase of the embryo (day), reaching the maximum at 24th day and then stabilized. Immunoblotting results confirmed that the expression of AQP-7 in the kidney gradually increased, reaching a peak after P24 d stabilized. Conclusion The expression of AQP-7 in the development of mouse renal tubules is spatiotemporal and plays an important role in the regulation of water and glycerol balance in the developing kidney.
