以水稻抗白叶枯病3对近等基因系(IR24与IRBB3、IR24与IRBB4和IR24与IRBB5)为材料,对近等基因系基因组DNA进行RAPD分析,结果显示:经120个10-核苷酸随机引物对3对近等基因系基因组DNA的PCR扩增,有2个引物(OPK-11和OPN-04)在IRBB3中各扩增出1~2条特异性条带;有3个引物(OPK-08、OPN-11和OPN-18)在IRBB4中各扩增出了2条特异性条带;有3个引物(OPL-04、OPN-10和OPO-18)在IRBB5中各扩增出1~2条特异性条带;在轮回亲本中未见有上述类似的片段被扩增,多次重复均获得较一致的结果。因此,推测这些特异性条带(DNA片段)分别与水稻抗白叶枯病抗性基因Xa-3、Xa-4和xa-5相关。
Near isogenic lines (IR24 and IRBB3, IR24 and IRBB4 and IR24 and IRBB5) were used as materials in RAPD analysis of rice isolates from rice bacterial blight. The results showed that after 120 10-nucleosides Two primers (OPK-11 and OPN-04) were used to amplify 1 or 2 specific bands in IRBB3 with 3 random primers and 3 pairs of near-isogenic genomic DNA. Three primers (OPK-08, OPN-11 and OPN-18) amplified two specific bands in IRBB4. Three primers (OPL-04, OPN-10 and OPO-18) 1 or 2 specific bands were added; no similar fragment was amplified in the recurrent parent, and more repeated results obtained more consistent results. Therefore, we speculated that these specific bands (DNA fragments) were related to the resistance genes of rice bacterial blight Xa-3, Xa-4 and xa-5, respectively.
