目的 探讨组织双氢睾酮(DHT)结合能力在良性前列腺增生(BPH)病因中的作用。方法32例BPH患者的前列腺组织取自耻骨上经膀胱前列腺摘除术,制备组织胞浆、胞核提取液,以乙醚萃取的方法去除提取液中所有的内源性类固醇激素,加入3H-DHT测定结合的3H -DHT的含量。结果 BPH组织中DHT结合蛋白的总DHT结合容量均数约为0.024nmol/g湿组织,胞浆、胞核DHT结合容量分别为(0.01280.0020)nmol/g湿组织和(0.01120.0059)nmol/g湿组织,两者比较差异无显著性(P>0.05)。结论 BPH组织中的DHT结合蛋白具有较高的DHT结合容量,维持局部DHT 的高浓度,有利于DHT在前列腺组织增生过程中的作用。
Objective To investigate the role of histone dihydrotestosterone (DHT) binding in the pathogenesis of benign prostatic hyperplasia (BPH). Methods Thirty-two cases of BPH were obtained from the suprapubic transvesical prostatectomy. The tissue cytoplasm and nucleus extract were prepared. All the endogenous steroid hormones in the extract were removed by ether extraction. 3H-DHT assay Bound 3H-DHT content. Results The total DHT binding capacity of DHT-binding protein in BPH tissues was about 0.024nmol / g wet tissue, and the DHT binding capacity in cytoplasm and nucleus were (0.01280.0020) nmol / g wet tissue and (0.01120.0059) nmol / g wet tissue, there was no significant difference between the two (P> 0.05). Conclusion The DHT-binding protein in BPH tissue has high DHT binding capacity and maintains the high concentration of local DHT, which is beneficial to the role of DHT in the proliferation of prostatic tissue.
