通过提取富硒女贞子蛋白和普通女贞子蛋白,采用MTT和ELISA法检测其对奶牛外周血淋巴细胞(peripheral blood lymphocyte;PBL)增殖活性和多种细胞因子分泌功能的影响。试验结果表明,2种蛋白均可促进ConA刺激下的奶牛外周血淋巴细胞增殖,效果随浓度的增加而上升,在浓度高于400 mg/L时普通女贞子蛋白较对照组效果显著(P<0.05),富硒女贞子蛋白差异极显著(P<0.01),且富硒女贞子蛋白在各个浓度均优于普通女贞子蛋白,差异显著(P<0.05)。富硒女贞子蛋白组的IL-2、IL4、IL-6、IFN-γ水平均显著高于对照组(P<0.05),其中IL-4水平与对照组差异极显著(P<0.01),普通女贞子蛋白的IL-4和IFN-γ较对照组也有显著提高(P<0.05)说明富硒女贞子蛋白可以进一步提高奶牛外周血淋巴细胞细胞活性,并调升细胞因子的分泌。
The effects of selenium-enriched Ligustrum lucidum protein and Ligustrum lucidum protein on the proliferation activity and the secretion of various cytokines in peripheral blood lymphocytes (PBL) of dairy cows were detected by MTT and ELISA. The results showed that both of the two proteins could promote the proliferation of peripheral blood lymphocytes of ConA stimulated by ConA, and the effect increased with the increase of concentration, and the effect of common Ligustrum lucidum protein was significant at the concentration higher than 400 mg / L (P (P <0.05). The protein of selenium-rich Ligustrum lucidum was significantly different (P <0.01), and the protein of selenium-rich Ligustrum lucidum was better than that of common Ligustrum lucidum in all concentrations (P <0.05). The levels of IL-2, IL4, IL-6 and IFN-γ in selenium-enriched Ligustrum lucidum protein group were significantly higher than those in control group (P <0.05). The level of IL-4 was significantly different from that in control group (P <0.05). The results showed that the selenium-enriched Ligustrum lucidum protein can further increase the cell activity of peripheral blood lymphocytes in cows and increase the secretion of cytokines .
