应用淋巴细胞杂交瘤技术,以H血型物质为抗原,经聚乙二醇(PEG)细胞融合,获得一株分泌IgG类抗人H血型抗原的单克隆抗体杂交瘤2G_6-H_8,细胞培养上清液效价1:128,亲和力10秒。经血凝特异试验、血凝抑制试验、吸收放散试验及临床验证,表明2G_6-H_8,单抗对人H血型抗原特异凝集,主要为抗O血型单抗。杂交瘤细胞置液氮中贮存一年,复苏传代后分泌抗体能力不变。单抗置56℃水浴30分钟,-20℃和37℃反复冻融三次,2~8℃保存半年,血凝特性不变。临床验证2505例,证明可替代传统抗H试剂,用做临床血型定型试剂之一。
