采用滤纸片法和打孔法2种抗菌试验方法,考察2-羟基-1-萘醛缩氨基脲(NAHSEM)对金黄色葡萄球菌、大肠杆菌、枯草杆菌的抗菌活性的结果表明,NAHSEM对3种细菌具有较强的抗菌活性。荧光光谱法研究了NAHSEM与牛血清白蛋白(BSA)的结合作用表明,这种结合行为使BSA的内源荧光猝灭。通过猝灭常数、结合常数和结合位点数的计算,证明荧光猝灭机理符合静态机制;NAHSEM和BSA形成1∶1稳定复合物。考察不同温度和酸度下的猝灭作用,进一步证实结合作用主要受范德华力和氢键驱动。
The antibacterial activity of 2-hydroxy-1-naphthylaldehyde semicarbazone (NAHSEM) against Staphylococcus aureus, Escherichia coli and Bacillus subtilis was tested by the two methods of antimicrobial test using filter paper and punching method. Bacteria have strong antibacterial activity. Fluorescence spectroscopy study of the binding of NAHSEM to bovine serum albumin (BSA) showed that this binding behavior quenched the endogenous fluorescence of BSA. The quenching constants, binding constants and the number of binding sites were calculated to show that the fluorescence quenching mechanism is consistent with the static mechanism; NAHSEM and BSA form a 1: 1 stable complex. The quenching effect under different temperature and acidity was investigated. It was further confirmed that the binding was mainly driven by van der Waals forces and hydrogen bonding.
