目的:通过研究肝纤维化大鼠原代肝星状细胞锌指结构9(ZF9)、结缔组织生长因子(CTGF)的表达及其动态变化,初步探讨CTGF、ZF9在肝星状细胞活化中的作用。方法:32只雄性SD大鼠皮下注射40%植物油CCl4,每周2次,并分别于注射1,4,8周后作大鼠肝原位灌注,分离肝星状细胞,Western blot法检测CTGF蛋白,RT-PCR检测ZF9 mRNA水平。结果:正常大鼠肝星状细胞ZF9基因可见弱表达,注射CCl41周后其表达增强,注射CCl44周后转录明显增强,8周时仍高表达。正常大鼠肝星状细胞CTGF蛋白无明显表达,模型组大鼠皮下注射CCl41周后,蛋白出现少量表达,注射CCl44周后,表达进一步增加,第8周CTGF蛋白表达更强。结论:ZF9、CT-GF基因随着肝损伤的加重,在肝星状细胞中的表达逐渐增加,共同促进肝纤维化的发生。
OBJECTIVE: To investigate the expression and dynamic changes of ZF9 and CTGF in primary hepatic stellate cells of rats with hepatic fibrosis and to explore the possible role of CTGF and ZF9 in the activation of hepatic stellate cells effect. Methods: Thirty-two male Sprague-Dawley rats were injected subcutaneously with 40% vegetable oil CCl4 twice a week for 1, 4 and 8 weeks after injection. Rat hepatic stellate cells were isolated and hepatic stellate cells were isolated. CTGF Protein, RT-PCR detection of ZF9 mRNA levels. Results: The expression of ZF9 gene in normal rat hepatic stellate cells was weakly expressed. The expression of ZF9 gene was enhanced after 41 weeks of CCl4 injection. The expression of ZF9 gene increased significantly after 44 weeks of CCl4 injection and was still high at 8 weeks. The expression of CTGF protein in normal rat hepatic stellate cells was not significantly increased. The expression of CTGF protein in rat hepatic stellate cells after 41 weeks of subcutaneous injection of CCl was slightly increased. The expression of CTGF protein was further increased after 44 weeks of CCl injection. Conclusion: The expression of ZF9 and CT-GF gene in hepatic stellate cells gradually increases along with the aggravation of hepatic injury, which can promote the occurrence of hepatic fibrosis.
