目的观察大鼠脑缺血再灌注后神经细胞巢蛋白(Nestin)、干细胞因子(SCF)和神经细胞黏附分子(NCAM)基因的表达。方法成年健康雌性SD大鼠36只,以线栓法建立大脑中动脉缺血再灌注模型,随机分为缺血1.5 h再灌注2 h、6 h、12 h、24 h、2 d、3 d、7 d、14 d组和假手术组,每组4只。应用原位杂交技术检测脑缺血再灌注后脑组织Nestin、SCF和NCAM mRNA的表达。结果假手术组皮质和纹状体区Nestin、SCF和NCAMmRNA均有微弱表达。脑缺血再灌注后Nestin mRNA表达增高,皮质除再灌注2 h以外、纹状体除再灌注2、6 h以外,其余各时间点均明显高于假手术组。SCF mRNA表达,皮质除再灌注2、6、12 h以外,纹状体除再灌注2、6 h以外,其余各时间点均明显高于假手术组。NCAM mRNA于再灌注2 h后在皮质和纹状体区开始表达,分别于再灌注12 h和1 d达高峰,7 d后逐渐减少,14 d降至假手术组水平。结论脑缺血再灌注后SCF mRNA表达可能具有促进神经干细胞增殖作用,NCAM表达可能参与了损伤后脑组织的修复过程。
Objective To observe the expression of Nestin, Stem Cell Factor (SCF) and Nerve Cell Adhesion Molecule (NCAM) in rats after cerebral ischemia-reperfusion. Methods Thirty-six adult female Sprague-Dawley rats were randomly divided into ischemia reperfusion (1.5 h), reperfusion (2 h, 6 h, 12 h, 24 h, 2 d, 3 d , 7 d, 14 d group and sham operation group, 4 rats in each group. In situ hybridization was used to detect the expression of Nestin, SCF and NCAM mRNA in rat brain after cerebral ischemia and reperfusion. Results Nestin, SCF and NCAM mRNA in the cortex and striatum of the sham group were all weakly expressed. The expression of Nestin mRNA increased after cerebral ischemia and reperfusion, except for 2 h after reperfusion, the striatum was significantly higher than sham group except for 2, 6 h after reperfusion. SCF mRNA expression, cortex in addition to 2, 6, 12 h after reperfusion, the striatum in addition to reperfusion after 2, 6 h, the rest of the time points were significantly higher than the sham group. NCAM mRNA began to express in cortex and striatum at 2 h after reperfusion, reached the peak at 12 h and 1 d after reperfusion, decreased gradually after 7 d, and dropped to the level of sham operation on the 14 th day. Conclusion The expression of SCF mRNA after cerebral ischemia-reperfusion may promote the proliferation of neural stem cells. The expression of NCAM may be involved in the repair process of brain tissue after injury.
