目的探讨端粒酶hTRT mRNA在口腔鳞癌及癌前病变组织中的表达相关性。方法采用原位杂交法检测病变组织中的hTRT mRNA的表达水平,标本取自哈尔滨医科大学第二附属医院口腔颌面外科病理室1980年至2000年的病理标本,其中口腔鳞癌46例、非典型增生18例、单纯性增生5例、肿瘤周围邻近组织5例、正常口腔黏膜2例,共76例口腔病理标本。结果80.5%鳞癌(37/46)、55.6%非典型增生(10/18)、20%单纯性增生(1/5)、20%肿瘤周围邻近组织(1/5)、0%正常口腔黏膜(0/2),端粒酶hTRTmRNA显示阳性表达。其中口腔鳞癌与非典型增生之间端粒酶hTRTmRNA表达差异有统计学意义(P<0.05),重度非典型增生与轻度及中度非典型增生之间端粒酶hTRT mRNA表达差异有统计学意义(P<0.05),口腔鳞癌各临床分期之间差异无统计学意义(P>0.5)。结论端粒酶hTRTmRNA在口腔鳞癌及癌前病变中均有表达,端粒酶活性的激活可能发生在口腔癌前病变晚期,其活性的激活是口腔癌前病变恶变及恶性肿瘤形成的重要因素。
Objective To investigate the expression of telomerase hTRT mRNA in oral squamous cell carcinoma and precancerous lesions. Methods The expression of hTRT mRNA in diseased tissues was detected by in situ hybridization. The specimens were collected from pathological specimens of oral and maxillofacial surgical pathology in Second Affiliated Hospital of Harbin Medical University from 1980 to 2000, including 46 cases of oral squamous cell carcinoma, 18 cases of hyperplasia, 5 cases of simple hyperplasia, 5 cases of adjacent tissue around the tumor, 2 cases of normal oral mucosa, a total of 76 cases of oral pathological specimens. Results 80.5% of squamous cell carcinomas (37/46), 55.6% of atypical hyperplasia (10/18), 20% of simple hyperplasia (1/5), 20% of adjacent tissues around the tumor (1/5), 0% of normal oral mucosa (0/2), telomerase hTRT mRNA showed positive expression. The expression of hTRTmRNA in telomerase between oral squamous cell carcinoma and atypical hyperplasia had statistical significance (P <0.05), there was statistical difference between hTRT mRNA expression between severe atypical hyperplasia and mild and moderate atypical hyperplasia (P <0.05). There was no significant difference in clinical staging between oral squamous cell carcinoma (P> 0.05). Conclusion Telomerase hTRT mRNA is expressed in both oral squamous cell carcinomas and precancerous lesions. The activation of telomerase activity may occur in the late stages of oral precancerous lesions. The activation of telomerase activity may be an important factor in the malignant transformation of oral precancerous lesions and malignant tumors .
