目的:探讨不同干燥方法对“祛湿化瘀方”防治实验性脂肪肝药效学的影响。方法:祛湿化瘀方中5味药物分别醇提、水提醇沉后合并,制备成生药含量为0.93 g.mL-1的药液,再按不同干燥方法制得浸膏、70℃真空负压干燥药物、40℃真空负压干燥药物、冷冻干燥药物、喷雾干燥药物。采用Wistar雄性大鼠,以四氯化碳(CCl4)复合高脂低蛋白饮食制备大鼠脂肪肝模型,造模2周后,将造模大鼠随机分为模型组、未干燥组、浸膏组、70℃真空负压干燥组、40℃真空负压干燥组、冷冻干燥组、喷雾干燥组,每组灌以相应药物2周。取材后检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、谷氨酰转移酶(GGT)活性、肝组织甘油三酯(TG)、游离脂肪酸(FFA)的含量变化。并对各个药物样品进行指纹图谱分析。结果:与正常组相比,模型组ALT,AST,GGT活性,肝组织TG,FFA含量显著升高。未干燥组与模型组相比,血清ALT,AST,GGT及肝组织TG,FFA显著下降;浸膏组与模型组相比,血清GGT,ALT,肝组织TG,FFA显著下降;70℃和40℃真空干燥组除血清ALT活性显著降低外,其余指标与模型组相比均无显著差异;冷冻干燥组除肝组织TG显著低于模型组,其余指标与模型组相比均无显著差异;喷雾干燥组除血清GGT显著低于模型组,其余指标与模型组相比均无显著差异;未干燥组的综合疗效优于其他各组。运用指纹图谱对各组药物进行分析,色谱图有相应的谱峰信息综合表达有差异。结论:“袪湿化瘀方”经不同干燥方法后对实验性脂肪肝药效均有所下降,不同方法干燥后部分有效物质丢失,可能是导致药效差异的主要原因。
Objective: To investigate the effects of different drying methods on the pharmacodynamic effects of “Qushi Huayu Recipe” in preventing and treating experimental fatty liver. Methods: Five herbs of Qushi Huayu Recipe were separately extracted by alcohol and combined with water and alcohol precipitation to prepare a liquid medicine with the crude drug content of 0.93 g.mL-1, then the extract was prepared according to different drying methods, Vacuum drying drugs, vacuum drying at 40 ℃ drugs, freeze-dried drugs, spray-dried drugs. Two weeks later, the model rats were randomly divided into model group, undisturbed group, extract group Group, vacuum vacuum drying group at 70 ℃, vacuum vacuum drying group at 40 ℃, freeze-drying group and spray-drying group. Each group was given corresponding drug for 2 weeks. The contents of ALT, AST, GGT, TG and FFA in liver tissue were determined after the preparation. And fingerprinting analysis of each drug sample. Results: Compared with normal group, ALT, AST, GGT activity and TG and FFA contents in liver tissue were significantly increased in model group. Compared with model group, serum ALT, AST, GGT and TG, FFA of liver tissue in untreated group decreased significantly. Compared with model group, serum GGT, ALT, TG and FFA of liver tissue decreased significantly in extract group and 70 ℃ and 40 ℃ vacuum drying group in addition to the serum ALT activity was significantly reduced, the remaining indicators were not significantly different from the model group; freeze-dried except liver TG was significantly lower than the model group, the remaining indicators compared with the model group no significant difference; spray In addition to serum GGT in the dry group was significantly lower than the model group, the remaining indicators were no significant difference compared with the model group; not dry group was better than the other groups. Fingerprinting analysis of each group of drugs, the chromatogram has the corresponding peak information of the comprehensive expression of differences. Conclusion: The effect of “Dan Hu Huanyu Recipe” on experimental fatty liver decreased with different drying methods. The loss of some effective substances after drying by different methods may be the main reason leading to the difference of pharmacodynamics.
