目的研究亚低温对过氧化氢损伤后脑微血管内皮细胞核转录因子(NF-κB)表达的影响。方法分离并培养大鼠脑微血管内皮细胞,用过氧化氢损伤的方法建立脑出血后血肿周围炎症反应内皮细胞模型。将细胞分为实验组(ICH)和对照组(C)。ICH组分为过氧化氢损伤常温组(ICH+N)和过氧化氢损伤亚低温组(ICH+H),C组分为常温对照组(C+N)和亚低温对照组(C+H)。采用Western blot技术检测各组NF-κB的表达水平。结果实验组NF-κB的表达水平均高于各自对照组(P<0.01),ICH+N组蛋白表达水平高于ICH+H组(P<0.01)。结论亚低温可有效降低过氧化氢损伤的脑微血管内皮细胞NF-κB的表达,从而减轻血肿周围炎症反应,发挥其脑保护作用。
Objective To investigate the effects of mild hypothermia on the expression of nuclear factor kappa B (NF-κB) in brain microvascular endothelial cells after hydrogen peroxide injury. Methods Rat brain microvascular endothelial cells were isolated and cultured. Endothelial cell models of inflammatory reaction around the hematoma were established by hydrogen peroxide injury. Cells were divided into experimental group (ICH) and control group (C). ICH was induced by hydrogen peroxide in ICH + N and ICH + H groups, C in C + N group and C + H in hypothermia control group, ). Western blot was used to detect the expression of NF-κB in each group. Results The expression of NF-κB in experimental group was higher than that in control group (P <0.01). The protein expression level in ICH + N group was higher than that in ICH + H group (P <0.01). Conclusion Mild hypothermia can effectively reduce the expression of NF-κB induced by hydrogen peroxide in brain microvascular endothelial cells, and thus reduce the inflammatory reaction around the hematoma and exert its neuroprotective effect.
