目的明确胃癌差异基因表达谱中弱差异表达基因的敏感性和生物学意义;探讨胃癌弱差异表达基因TINF2在胃癌发生发展中的生物学作用和意义。方法利用GoMiner软件研究TINF2的生物学功能。并利用Real-Time PCR法以及Western blot法验证胃癌弱差异基因表达谱中TINF2基因的表达水平。结果弱差异表达基因TINF2定位于染色体端粒区,主要具有蛋白结合、端粒DNA结合活性,通过端粒酶调节端粒长度的功能;通过20对胃癌及配对的癌旁正常组织实验室验证,TINF2的mRNA及蛋白表达水平在胃癌组织中均表现为低表达(17/20,85%),差异具有显著性(P<0.001),与前期基因芯片中胃癌TINF2表达水平一致。结论 TINF2异常表达与胃癌的发生联系密切,并且可能在胃癌的发生发展中发挥重要的生物学作用。
Objective To clarify the sensitivity and biological significance of differentially expressed genes in differentially expressed genes in gastric cancer and to explore the biological roles and significance of differentially expressed genes TINF2 in gastric carcinogenesis. Methods The biological function of TINF2 was studied by using GoMiner software. Real-Time PCR and Western blot were used to verify the expression of TINF2 gene in the differentially expressed genes of gastric cancer. Results TINF2, a weakly differentially expressed gene, was located in the telomere region of the chromosome and mainly involved in protein binding and telomere DNA binding activity. Telomerase was used to regulate the telomere length. Twenty-two gastric cancer tissues and paired adjacent normal tissues were verified by laboratory tests. TINF2 mRNA and protein expression in gastric cancer showed low expression (17/20, 85%), the difference was significant (P <0.001), consistent with the previous gene chip TINF2 expression levels. Conclusion The abnormal expression of TINF2 is closely related to the occurrence of gastric cancer and may play an important biological role in the development of gastric cancer.
