沙棘水提物对短波紫外线诱导HaCaT细胞凋亡及线粒体膜电位的影响

目的:研究沙棘(Hippophae rhamnoides L.)水提物对短波紫外线(UVC)辐射人永生化角质形成(Ha Ca T)细胞增殖与凋亡、线粒体膜电位(ΔΨm)的影响,探讨沙棘水提物对UVC辐射Ha Ca T细胞是否具有防护功能及其作用机制。方法:体外培养Ha Ca T细胞株,设置对照组(不加药、不照射)、UVC组和药物组(UVC辐射+不同浓度的沙棘水提物),CCK-8法测定细胞增殖、Annexin V/PI流式细胞术检测细胞凋亡、JC-1检测ΔΨm。结果:与UVC组比较,药物组CCK-8吸光度值(OD值)明显升高(P<0.01),且呈剂量依赖;细胞凋亡率降低,发生线粒体膜电位下降的细胞数比例(F值)下降。结论:沙棘水提物能够抑制Ha Ca T细胞凋亡、促进细胞增殖,其机制可能与保护线粒体功能有关。 OBJECTIVE: To study the effects of Hippophae rhamnoides L. extract on proliferation, apoptosis and mitochondrial membrane potential (ΔΨm) of human keratinocytes (HaCa T) irradiated by shortwave ultraviolet (UVC) radiation, Whether UVC radiation HaCa T cells have protective function and its mechanism. Methods: HaCa T cells were cultured in vitro. The control group (without drug and no irradiation), UVC group and drug group (UVC radiation + water extract of sea buckthorn at different concentrations) were set up. The cell proliferation was measured by CCK- / PI flow cytometry detection of apoptosis, JC-1 detection ΔΨm. Results: Compared with UVC group, CCK-8 absorbance value (OD value) in drug group was significantly increased (P <0.01), and in a dose-dependent manner; the apoptosis rate decreased, the proportion of cells with mitochondrial membrane potential decreased )decline. Conclusion: The aqueous extract of seabuckthorn can inhibit the apoptosis of HaCa T cells and promote cell proliferation, and its mechanism may be related to the protection of mitochondrial function.
