目的:研究小鼠胸膜间皮细胞免疫相关基因表达谱的状况,从而探索用现代免疫及基因芯片技术评价解释中医肺气学说的新方法。方法:取8只BALB/c小鼠胸膜组织块剪切成碎块,提取mRNA,经标记、杂交、清洗、芯片扫描、图像采集与数据分析等过程获得基因表达谱。结果:共筛选出小鼠胸膜间皮细胞53条免疫相关高表达基因。其中主要有免疫应答高表达基因14条;炎性反应高表达基因6条;非特异性免疫应答高表达基因7条;免疫应答调节高表达基因4条;急性反应阶段高表达基因3条;抗原加工与提呈高表达基因2条;Ⅱ型超敏反应正向调节高表达基因2条。结论:从现代免疫学观点及正常小鼠胸膜间皮细胞基因芯片的研究来看,胸膜间皮细胞既表达免疫相关的正向调节基因,亦表达负向免疫相关的调节基因。这不仅可为现代免疫学增添新的内容,而且对丰富中医肺气学说,研究中医肺脏的实质乃至推动中医学理论的深入研究都将具有积极意义,为胸膜的其他后续研究构建一平台,为丰富中医肺气学说与肺气的实质提供分子生物学的基础研究依据;弥补这方面的研究空白。
OBJECTIVE: To study the expression profile of immune-related genes in mouse pleural mesothelial cells, and to explore a new method to evaluate the interpretation of TCM pneumoconiosis using modern immunology and gene chip technology. Methods: Eight BALB / c mice were cut into pieces and cut into pieces. The mRNA expression was obtained by labeling, hybridization, washing, chip scanning, image acquisition and data analysis. Results: A total of 53 immune-related high expression genes of mouse pleural mesothelial cells were screened out. Among them, there are 14 genes highly expressed in immune response, 6 genes highly expressed in inflammatory reaction, 7 genes highly expressed in nonspecific immune response, 4 genes highly regulated in immune response, 3 genes highly expressed in acute phase, And presented high expression of two genes; type Ⅱ hypersensitivity positive regulation of two highly expressed genes. Conclusion: From the perspective of modern immunology and normal mouse pleural mesothelial cell microarray research, pleural mesothelial cells express both immune-related positive regulatory genes and negative immune-related regulatory genes. This will not only add new content to modern immunology, but also enrich the TCM theory of lung qi, study the essence of Chinese medicine lung and even promote in-depth study of Chinese medicine theory will have a positive meaning for the other follow-up study of pleural to build a platform for Enrich the essence of TCM theory of lung qi and lung qi to provide basic research basis of molecular biology; make up the blank of this research.
