[目的]探讨DDB2基因单核苷酸多态性与焦炉工外周血淋巴细胞DNA损伤的关联。[方法]选取焦化厂在岗焦炉工325名作为接触组,选择同厂无职业多环芳烃暴露者150人作为对照组,使用反向高效液相色谱法测定尿1-羟基芘含量,采用碱性单细胞凝胶电泳技术检测淋巴细胞DNA损伤情况,应用TaqMan探针荧光标记聚合酶链反应方法对DDB2基因进行分型。[结果]接触组尿1-羟基芘含量[(4.41±1.06)μmol/mol肌酐]高于对照组[(3.33±1.03)μmol/mol肌酐],差异有统计学意义(P<0.05);接触组外周血淋巴细胞彗星尾距(olive tail moment,OTM)[0.41(0.08~4.28)]也高于对照组[0.31(0.08~3.16)],差异有统计学意义(P<0.05)。两组中DDB2基因SNPs的不同基因型携带者OTM差异均无统计学意义。分层结果表明,对照组年龄<39岁个体中,启动子区rs2029298位点AA基因型携带者DNA损伤高于GG基因型携带者,差异有统计学意义(P<0.05);其他差异均无统计学意义。[结论]DDB2基因多态性对焦炉工外周血淋巴细胞DNA损伤程度无影响,在对照组低年龄个体中rs2029298位点AA基因型携带者的DNA损伤程度高于GG基因型携带者。
[Objective] To investigate the association between single nucleotide polymorphism of DDB2 gene and DNA damage in peripheral blood lymphocytes of coke oven workers. [Method] A total of 325 coke oven workers in the coke plant were selected as the contact group. 150 workers without occupational PAHs were selected as the control group. The urine 1-hydroxypyrene content was determined by reverse-phase high performance liquid chromatography. The single cell gel electrophoresis was used to detect the DNA damage of lymphocytes. The TaqMan probe was used to detect the DDB2 gene by fluorescence-labeled polymerase chain reaction. [Result] Compared with control group [(3.33 ± 1.03) μmol / mol creatinine], the content of urinary 1-hydroxypyrene in contact group was significantly higher than that of control group [(4.41 ± 1.06) μmol / mol creatinine] The values of OPT (0.41 (0.08 ~ 4.28)] in peripheral blood lymphocytes of peripheral blood were also higher than that of control group [0.31 (0.08 ~ 3.16)], the difference was statistically significant (P <0.05). There was no significant difference in OTM among different genotype carriers of DDB2 gene SNPs in both groups. The results of stratification showed that the DNA damage of AA genotypes in rs2029298 promoter region was higher in the control group than in the genotype GG carriers (P <0.05), but there was no significant difference in other groups Statistical significance. [Conclusion] The polymorphism of DDB2 gene has no effect on the DNA damage of peripheral blood lymphocytes in coke oven workers. The DNA damage of AA genotype rs2029298 locus in low-age individuals of control group is higher than that of GG genotype carriers.
