本研究以牛中性粒细胞牛溶菌肽的线性衍生物Bac2a为研究对象,以蜂毒素为对照肽,探讨Bac2a的生物特性及其稳定性。采用圆二色谱法、微量稀释法、噻唑蓝(MTT)法等方法测定了Bac2a的二级结构、抑菌活性、溶血活性、细胞毒性及其稳定性。研究结果表明:Bac2a在水溶液中呈无规则卷曲状,在疏水环境中则呈α-螺旋结构;Bac2a的抑菌作用较强,最小抑菌浓度(MIC)为2~32μmol/L;当肽浓度为64μmol/L时,其溶血率较低,仅为14.81%;细胞毒性小,治疗指数为3.26,细胞选择性较强;100℃处理1 h后,Bac2a依然保持较强的抑菌活性,热稳定性好。因此,Bac2a在食品、医药和其它领域都具有良好的应用前景,有望成为抗生素替代品。
In this study, Bac2a, a linear derivative of bovine neutrophil bovine lysozyme, was used as a study object. The melittin was used as a control peptide to investigate the biological characteristics and the stability of Bac2a. The secondary structure, antibacterial activity, hemolytic activity, cytotoxicity and stability of Bac2a were determined by circular dichroism method, microdilution method and MTT method. The results showed that Bac2a showed irregular curly shape in aqueous solution and α-helical structure in hydrophobic environment. Bac2a had strong antibacterial activity with the MIC of 2 ~ 32μmol / L. When Bac2a concentration At 64μmol / L, the hemolysis rate was low, only 14.81%; the cytotoxicity was small, the therapeutic index was 3.26, the cell selectivity was strong; Bac2a still maintained strong antibacterial activity after 100 ℃ treatment for 1 h, Good stability. Therefore, Bac2a has good application prospects in food, medicine and other fields and is expected to become an antibiotic substitute.
