目的:研究黑布药膏对兔耳增生性瘢痕Ⅰ,Ⅲ胶原及基质金属蛋白酶-1 mRAN表达的影响。方法:成年大耳白兔24只,随机分为正常对照组、瘢痕模型组和黑布药膏治疗组。瘢痕模型组和黑布药膏治疗组建立兔耳腹侧面增生性瘢痕动物模型,21 d后,造模成功。在黑布药膏治疗组瘢痕局部和正常对照组对应皮肤涂抹黑布药膏,每3 d 1次,每次1 g,连续用药56d。在用药后第2,4,6,8周分别切取两组瘢痕组织,通过实时定量PCR(RT-PCR)法检测Ⅰ,Ⅲ胶原mRAN及基质金属蛋白酶-1 mRAN的表达。结果:瘢痕模型组与正常对照组比较Ⅰ胶原mRAN表达显著增强,Ⅲ胶原mRAN及基质金属蛋白酶-1 mRAN表达显著减弱(P<0.01)。黑布药膏治疗组在用药后第2,8周Ⅰ胶原mRAN表达分别为(3.62±1.70),(1.34±0.52);模型对照组第2,8周Ⅰ胶原mRAN表达分别为(3.15±1.24),(4.53±1.71),黑布药膏治疗组较模型组显著减弱(P<0.01);黑布药膏治疗组在用药后第2,8周Ⅲ胶原mRAN表达分别为(1.46±0.34),(2.59±0.35),模型对照组Ⅲ胶原mRAN表达分别为(0.88±0.14),(0.37±0.08),黑布药膏治疗组较模型组显著增强(P<0.01);黑布药膏治疗组在用药后第2,8周基质金属蛋白酶-1 mRAN表达分别为(1.77±0.40),(4.76±0.77),模型对照组第2,8周基质金属蛋白酶-1 mRAN表达分别为(0.53±0.14),(0.34±0.04),黑布药膏治疗组较模型组显著增强(P<0.01)。结论:黑布药膏可以通过抑制I型胶原mRNA的表达,促进Ⅲ型胶原、基质金属蛋白酶-1mRNA的表达,增加胶原酶含量,降低瘢痕组织胶原含量,从而抑制兔耳增生性瘢痕组织增生,这可能是黑布药膏抑制兔耳增生性瘢痕增生的机制之一。
Objective: To study the effect of black cloth ointment on mRAN expression of collagen Ⅰ and Ⅲ and matrix metalloproteinase-1 in hypertrophic scar of rabbit ear. Methods: Twenty-four adult rabbits were randomly divided into normal control group, scars model group and black cloth ointment treatment group. The scars model group and the black cloth ointment treatment group established rabbit ear ventral hyperplastic scar animal model, 21 days later, the model was successful. In the black cloth ointment treatment group local scar and normal control group corresponding to the skin smear black cloth ointment, 1 d every 3 days, each 1 g, continuous medication 56d. At 2, 4, 6 and 8 weeks after operation, two groups of scar tissue were excised, and the expression of mRAN and MMP-1 mRAN were detected by real-time quantitative PCR (RT- Results: Compared with the normal control group, the expression of mRAN in collagen Ⅰ group was significantly increased, and the expression of mRAN Ⅲ and mRAN in collagen Ⅲ significantly decreased (P <0.01). The mRNA expression of mRAN in the black ointment treated group was (3.62 ± 1.70) and (1.34 ± 0.52) respectively at the second and eighth week after treatment. The expression of mRAN at the second and eighth week and the first and eighth week in the model control group were (3.15 ± 1.24) , (4.53 ± 1.71) respectively. Compared with the model group, the cupping treatment group was significantly weaker (P <0.01). The mRNA expression of mRAN in the black ointment group was (1.46 ± 0.34), (2.59 ± 0.35). The mRNA expression of mRAN in model group Ⅲ was (0.88 ± 0.14) and (0.37 ± 0.08) respectively, and the black cloth ointment treatment group was significantly higher than the model group (P <0.01). The black cloth ointment treatment group The mRNA expression of MMP-1 at 2 and 8 weeks were (1.77 ± 0.40) and (4.76 ± 0.77), respectively. Conclusion: The black cloth ointment can inhibit the proliferation of hypertrophic scar tissue in rabbit ear by inhibiting the expression of type I collagen mRNA, promoting the expression of type Ⅲ collagen and matrix metalloproteinase-1 mRNA, increasing the content of collagenase and reducing the collagen content in scar tissue It may be one of the mechanisms by which the black cloth ointment suppresses the hyperplasia of rabbit ear hyperplasia.
