目的研究PINCH-1因子在急性白血病骨髓基质细胞中的表达及意义。方法培养骨髓基质细胞,用West-ern-blotting和RT-PCR观察PINCH-1 mRNA和蛋白的表达情况;进行免疫细胞化学和免疫荧光染色,观察PINCH-1蛋白的表达及其在细胞内的分布情况。结果在急性淋巴细胞白血病和急性髓系白血病患者骨髓基质细胞中,PINCH-1在mRNA水平和蛋白水平均比正常骨髓基质细胞增高(P<0.01);急性淋巴白血病与急性髓系白血病之间PINCH-1蛋白的表达有显著差异(P<0.01),但mRNA表达无明显差别。结论PINCH-1在急性白血病骨髓基质细胞表达明显高于正常骨髓基质细胞,可能与急性白血病发生发展相关。
Objective To study the expression of PINCH-1 in bone marrow stromal cells of acute leukemia and its significance. Methods Bone marrow stromal cells were cultured and the expression of PINCH-1 mRNA and protein was observed by West-ern-blotting and RT-PCR. Immunocytochemistry and immunofluorescence staining were used to observe the expression of PINCH-1 protein and its distribution within the cells Happening. Results In both acute lymphoblastic leukemia and acute myeloid leukemia patients, PINCH-1 mRNA and protein levels in bone marrow stromal cells were significantly higher than those in normal bone marrow stromal cells (P <0.01). Acute lymphoblastic leukemia and acute myeloid leukemia PINCH -1 protein expression was significantly different (P <0.01), but no significant difference in mRNA expression. Conclusion The expression of PINCH-1 in acute leukemia bone marrow stromal cells is significantly higher than that of normal bone marrow stromal cells, which may be related to the development of acute leukemia.
