目的观察软肝化坚颗粒对肝纤维化模型C57小鼠肝组织中α-平滑肌肌动蛋白(α-SMA)、转化生长因子(TGF)β1表达和I型胶原水平的影响。方法 40只C57小鼠随机分为4组,分别为正常对照组、模型对照组、软肝化坚颗粒低剂量组及高剂量组,每组10只。采用四氯化碳(CCl4)腹腔注射制造小鼠肝纤维化模型,通过免疫组织化学染色方法检测肝组织中α-SMA、TGFβ1和I型胶原的表达,同时进行肝组织纤维化评分(S)。结果各模型组肝组织α-SMA、TGFβ1及Ⅰ型胶原表达和纤维化评分与正常对照组比较,差异均具有统计学意义(P<0.05);软肝化坚颗粒低剂量组、高剂量组肝组织α-SMA和TGFβ1及Ⅰ型胶原评分和纤维化评分较模型对照组均明显降低,差异具有统计学意义(P<0.05);软肝化坚颗粒低剂量组和高剂量组α-SMA、TGFβ1及Ⅰ型胶原评分及肝纤维化评分之间差异无统计学意义(P>0.05)。结论软肝化坚颗粒可能通过抑制C57小鼠肝组织TGFβ1的产生,进而抑制肝星状细胞(HSC)的活化,减少细胞外基质(ECM)的合成,从而降低I型胶原增生,达到预防和治疗肝纤维化的作用。
Objective To observe the effect of Ruanghujianjian Granule on the expression of α-smooth muscle actin (α-SMA), transforming growth factor (TGFβ1) and type I collagen in liver of C57 mice with hepatic fibrosis. Methods Forty C57 mice were randomly divided into 4 groups: normal control group, model control group, low-dose soft-drink preparation and high-dose group, with 10 rats in each group. The model of hepatic fibrosis was induced by intraperitoneal injection of carbon tetrachloride (CCl4). The expression of α-SMA, TGFβ1 and type I collagen in liver tissue was detected by immunohistochemical staining, and the liver fibrosis score (S) . Results The expression of α-SMA, TGFβ1, collagen type Ⅰ and fibrosis scores of liver tissue in each model group were significantly lower than those in normal control group (P <0.05) Α-SMA, TGFβ1, collagen type Ⅰ and fibrosis score of liver tissue in model group were significantly lower than those in model control group (P <0.05) , TGFβ1 and type Ⅰ collagen score and liver fibrosis score was no significant difference (P> 0.05). Conclusion Rouganhuijian granule could inhibit the activation of hepatic stellate cells (HSC), decrease the synthesis of extracellular matrix (ECM), and reduce type I collagen hyperplasia by inhibiting the production of TGFβ1 in liver of C57 mice, Therapeutic effect of liver fibrosis.
