Objective:To investigate the effects of raloxifene and estradiol benzoate on expression of estrogen sulfotransferase (EST) in normal human breast tissue implanted into nude mice. Methods:Fifteen specimens of normal human breast tissue from patients with benign breast disease were implanted into 9-10-week-old athymic nude mice.Every specimen was divided into five parts and individually implanted into five mice.The mice were divided into 5 groups subjected to the following treatments:distilled water(Control, GroupⅠ);estradiol benzoate(B-E_2) at 1 mg/kg(GroupⅡ);raloxifene at 0.3 mg/kg(GroupⅢ), 3 mg/kg(GroupⅣ) and 30 mg/kg(GroupⅤ).Treatment was given every day,and human breast tissues were removed for experiment after treatment for 30 days.The expression of EST mRNA was detected by reverse transcription polymerase chain reaction(RT-PCR). Results:The semi-quantitative EST mRNA was increased by about twofold in breast tissue in estradiol benzoate group compared to that of control(P<0.05).But there was no significant difference between any group of raloxifene and the control(P>0.05). Conclusion:Dose of 1 mg/kg estradiol benzoate increases the EST expression,while raloxifene has no effect on expression of EST in normal breast tissue.
