目的 :探讨隐睾中诱生型一氧化氮合成酶 (i NOS)基因表达与生殖细胞凋亡的关系。方法 :选取日龄为2 2 d的 SD雄性大鼠 2 0只 ,建立单侧隐睾模型 ,用生物素 - d U TP/酶标亲和素测定法检测睾丸生殖细胞凋亡 ,用免疫组织化学 SP法检测 i NOS基因表达。结果 :隐睾生殖细胞凋亡指数在术后 7d内随时间的延长而增加 ;从术后第 4天起 ,与对侧正常睾丸相比 ,隐睾发生凋亡的生殖细胞显著增加 (P <0 .0 1) ;在正常的睾丸组织中 ,i NOS定位于间质细胞、支持细胞和初级精母细胞的胞浆中 ,在隐睾精曲小管脱落的细胞胞浆中 ,i NOS免疫染色加强。结论 :生殖细胞凋亡增加 ,i NOS基因表达增强是生殖细胞凋亡增加的重要原因之一
Objective: To investigate the relationship between inducible nitric oxide synthase (iNOS) gene expression and germ cell apoptosis in cryptorchidism. Methods: Twenty male Sprague-Dawley rats of 22 days old were selected to establish unilateral cryptorchidism model. The apoptosis of testicular germ cells was detected by biotin-dUTP / Chemical SP method to detect iNOS gene expression. Results: The apoptosis index of cryptorchid germ cells increased with time within 7 days after operation. From the 4th day after operation, the germ cells apoptosis increased significantly in cryptorchid testis compared with the contralateral normal testis (P < 0.010). In normal testicular tissue, i NOS was localized in the cytoplasm of mesenchymal cells, supporting cells and primary spermatocytes, and i NOS immunostaining strengthen. Conclusion: The increase of germ cell apoptosis and the increase of iNOS gene expression are one of the important reasons for the increase of germ cell apoptosis
