目的探讨硫酸镍(NiSO4)对大鼠睾丸细胞能量代谢的影响。方法健康Wistar雄性大鼠40只,随机分为4组,即正常对照组(NS组),NiSO41.25、2.50和5.00 mg/kg,腹腔注射染毒,1次/d,连续30 d。石墨炉原子吸收光谱法检测大鼠睾丸组织镍含量;Western blot法测定琥珀酸脱氢酶(SDH)、苹果酸脱氢酶(MDH)和谷胱甘肽过氧化物酶(GSH-Px)蛋白表达;分光光度法测定大鼠睾丸细胞线粒体中SDH、MDH和GSH-Px活力以及睾丸组织中葡萄糖(GLU)和乳酸(LD)的含量。结果 NiSO42.50和5.00 mg/kg组睾丸中Ni含量高于NS组(P<0.05);2.50和5.00 mg/kg组SDH、各剂量组MDH和5.00 mg/kg组GSH-Px蛋白均表达下调(P<0.05);2.50和5.00 mg/kg组大鼠睾丸线粒体中MDH和SDH酶活力和睾丸组织中GLU含量低于NS组(P<0.05);5.00 mg/kg组大鼠睾丸线粒体中GSH-Px低于NS组(P<0.05)。结论 NiSO4致雄性大鼠睾丸细胞损伤可能与其干扰能量代谢有关。
Objective To investigate the effect of nickel sulfate (NiSO4) on the energy metabolism of testicular cells in rats. Methods Forty healthy Wistar male rats were randomly divided into 4 groups: normal control group (NS group), NiSO41.25, 2.50 and 5.00 mg / kg, intraperitoneal injection once a day for 30 days. Graphite furnace atomic absorption spectrometry was used to detect the content of nickel in testis tissue of rats. Western blot was used to determine the contents of succinate dehydrogenase (SDH), malate dehydrogenase (MDH) and glutathione peroxidase (GSH-Px) The activity of SDH, MDH and GSH-Px in rat testicular mitochondria and the content of glucose (GLU) and lactate (LD) in testis were determined by spectrophotometry. Results The contents of Ni in testis of NiSO42.50 and 5.00 mg / kg groups were significantly higher than those in NS group (P <0.05). The levels of GSH-Px protein in MDS of 2.50 and 5.00 mg / kg group and MDH of 5.00 mg / kg group were all decreased (P <0.05). The activities of MDH and SDH in testis mitochondria and the content of GLU in testes of 2.50 and 5.00 mg / kg groups were lower than those in NS group (P <0.05) -Px was lower than NS group (P <0.05). Conclusion The damage of testicular cells in male rats induced by NiSO4 may be related to the interference of energy metabolism.
