为研究昆明山海棠 (THH)的药理和毒理作用 ,通过流式细胞仪检测发现THH对Jurkat,CHE和NIH3T3细胞均有较强的致凋亡作用 ,但Jurkat淋巴瘤细胞更为敏感 ,每皿 5 0 μLTHH作用 48h后 ,可使 93%的肿瘤细胞凋亡 .使用Annexin V Fluos检测Jurkat细胞 ,结果发现THH作用 0 5h后即可出现早期细胞凋亡 (细胞膜损害 ) ,4~ 6h后 15 0~ 2 0 0 μLTHH组膜损伤率可高达 85 %,证明THH首先是损伤细胞膜 ,再发展为细胞凋亡的 .上述发现有助于阐明THH治疗自身免疫性疾病和潜在抗肿瘤的作用机理
In order to study the pharmacological and toxicological effects of THH, THH has a strong apoptosis-inducing effect on Jurkat, CHE and NIH3T3 cells by flow cytometry, but Jurkat lymphoma cells are more sensitive. After treatment with 50 μL THH for 48 h, 93% of the tumor cells were able to undergo apoptosis. Jurkat cells were detected using Annexin V Fluos. It was found that early apoptosis (cell membrane damage) occurred after THH 5 h, and 4 to 6 h later. The damage rate in the 0–200 μL THH group was as high as 85%, demonstrating that THH first damages the cell membrane and then develops apoptosis. These findings help elucidate the mechanism of THH in the treatment of autoimmune diseases and potential anti-tumor mechanisms.
