目的:研究大鼠胰腺β肿瘤细胞(INS-1)中,生长激素(GH)、催乳素(PRL)、葡萄糖和胰岛素对胰岛素样生长因子结合蛋白-2和3(IGFBP-2、3)的表达调控,探索IGFBP-2和3对维持正常β细胞生理功能的作用。方法:RNA印迹法检测INS-1细胞中IGFI3P-2和3的mRNA表达情况及蛋白免疫印迹法分析INS-1细胞中IGFBP-3和2蛋白表达。结果:在INS-1细胞中,GH和PRL调控IGFBP-3 mRNA和蛋白表达。葡萄糖对IGFBP-2 mRNA调节作用不同:高浓度(10 mmol/L)葡萄糖条件下,IGFBP-2 mRNA表达被抑制;低浓度(1.0~5.6 mmol/L)葡萄糖条件下,IGFBP-2 mRNA有表达。葡萄糖浓度为5.6 mmol/L时,加入GH孵育16 h,IGFBP-2 mRNA表达被抑制。外源胰岛素抑制 IGFBP-2 mRNA的表达。结论:GH和PRL可以调控IGFBP-3的mRNA和蛋白表达,IGFBP-2 mRNA表达主要通过胰岛素调控。
AIM: To investigate the effect of growth hormone (GH), prolactin (PRL), glucose and insulin on insulin-like growth factor binding protein-2 and 3 (IGFBP-2,3) Expression and regulation, to explore IGFBP-2 and 3 on the maintenance of normal β-cell physiological function. Methods: The mRNA expression of IGFI3P-2 and 3 in INS-1 cells was detected by Northern blotting and the protein expression of IGFBP-3 and 2 in INS-1 cells was analyzed by Western blotting. Results: In INS-1 cells, GH and PRL regulate IGFBP-3 mRNA and protein expression. The effect of glucose on the regulation of IGFBP-2 mRNA is different: the expression of IGFBP-2 mRNA is inhibited under high concentration of glucose (10 mmol / L); the expression of IGFBP-2 mRNA under low concentration (1.0-5.6 mmol / L) of glucose . When glucose concentration was 5.6 mmol / L, IGFBP-2 mRNA expression was inhibited by adding GH for 16 h. Exogenous insulin inhibits IGFBP-2 mRNA expression. Conclusion: GH and PRL can regulate IGFBP-3 mRNA and protein expression, and IGFBP-2 mRNA expression is mainly regulated by insulin.
