为了分析转基因显微注射的注射剂量对胚胎发育效果的影响,用尖端直径为0.5~1.0um的显微注射针在不同气压和不同时间的条件下进行微注射测试,并对133枚小鼠受精卵分别注射不同剂量,观察注射后胚胎发充情况。实验采用的气压电控式显微注射器,具有注射时间短(msec),注射的可控性和可重复性好的特点,在相同条件下重复注射时,误差小于0.004pl。结果表明,小鼠受精卵原核内注射剂量在2~4pl时,胚胎体外发育不受影响,当大于10pl时,胚胎在注射后1h内全部死亡,也证明了注射针尖端直径小于1.0um时,导致细胞损伤和影响胚胎发育的主要因素是注射剂量的大小。因此,对于小鼠胚胎原核内注射剂量2~4pl是较为合适的“安全剂量
In order to analyze the effects of injection dose of genetically modified microinjection on embryo development, microinjection test was performed under the condition of different pressure and different time with micro-injection needle with tip diameter of 0.5-1.0um, Mouse fertilized eggs were injected with different doses to observe the situation after injection of embryonic hair.
