本文介绍了粒系定向干细胞的体内扩散盒琼脂培养技术。将含0.5%琼脂的骨髓细胞悬液注入扩散盒,植入正常小鼠腹腔培养5天,细胞团生长良好。骨髓细胞种植量与细胞团产率之间呈正相关。体内扩散盒琼脂培养法无需提供外源性CSF,造血干细胞能在更接近干体内正常内环境的条件下增殖,是反映粒系定向干细胞相对数量的一个较好的实验技术。
This article describes in vivo diffusion cassette agar culture of granulocytic committed stem cells. The myeloid cell suspension containing 0.5% agar was injected into the diffusion box and implanted into normal mice for 5 days. The cell mass grew well. There was a positive correlation between the amount of bone marrow cells planted and the cell mass production rate. In vivo proliferation cassette agar culture does not need to provide exogenous CSF, and hematopoietic stem cells can proliferate more closely to normal inner environment of dry body, which is a good experimental technique to reflect the relative number of granule-derived stem cells.
