目的:研究丹参素对巨噬细胞的肝X受体(LXR)及其下游的目的基因的表达和胆固醇外流的影响,探讨丹参素对LXR信号系统的作用。方法:分离人外周血的单核细胞,并转化为巨噬细胞。在丹参素的作用下,观察巨噬细胞的aopA-I介导的胆固醇外流的变化和LXR及其下游目的基因的mRNA及蛋白的表达。结果:丹参素增强细胞内胆固醇的外流,使胆固醇外流的基因LXRα及其下游的一些目的基因表达上调,并呈剂量依赖性。结论:巨噬细胞在丹参素的作用下,上调LXR及其下游影响胆固醇代谢的目的基因,从而使胆固醇外流增强。
Objective: To investigate the effects of Danshensu on the expression of LXR and its downstream target genes and cholesterol efflux in macrophages, and to explore the effect of Danshensu on LXR signaling system. Methods: Human peripheral blood mononuclear cells were isolated and transformed into macrophages. Under the action of Danshensu, we observed the change of aopA-I-mediated cholesterol efflux and the mRNA and protein expression of LXR and its downstream target genes in macrophages. Results: Danshensu enhanced the efflux of intracellular cholesterol and up-regulated the expression of LXRα and some downstream genes of cholesterol efflux in a dose-dependent manner. Conclusion: Under the action of Danshensu, macrophages up-regulate LXR and its downstream target genes that affect cholesterol metabolism, leading to enhanced cholesterol efflux.
