目的:建立一种反相高效液相色谱分析方法,同时测定小柴胡汤中黄芩苷和甘草酸的含量。方法:采用Agilent Zorbax SB C_(18)柱(4.6mm×250mm,5m),以乙腈-0.01%磷酸溶液为流动相,梯度洗脱,流速1.0mL/min.柱温25℃,检测波长为250nm。结果:黄芩苷和甘草酸单铵盐在50min内即实现良好分离,黄芩苷浓度在12.66~405.0g/mL线性范围内呈良好的线性关系,其回归方程为:Y=19.726X+13.972,r= 0.9998。甘草酸在1.988~63.60g/mL的线性范围内呈良好的线性关系,其回归方程为:Y=10.765X+ 6.6433,r=0.9997。两种成分精密度试验RSD<1%;48h内稳定性试验RSD<1%;加样回收率试验RSD<2%。结论:该方法简便快速、准确可靠,能同时测定小柴胡汤中黄芩苷及甘草酸两种有效成分的含量,为有效控制小柴胡汤的质量提供参考。
Objective: To establish a reversed-phase high performance liquid chromatography method for the determination of baicalin and glycyrrhizic acid in Xiaochaihu decoction. METHODS: Agilent Zorbax SB C 18 column (4.6mm×250mm, 5m) was used. The mobile phase consisted of acetonitrile-0.01% phosphoric acid solution with gradient elution at a flow rate of 1.0mL/min. The column temperature was 25°C and the detection wavelength was 250 nm. Results: The baicalin and glycyrrhizic acid monoammonium salt achieved good separation in 50 min. The baicalin concentration showed a good linear relationship in the linear range of 12.66-405.0 g/mL. The regression equation was: Y=19.726X +13.972, r = 0.9998. Glycyrrhizic acid showed a good linear relationship in the linear range of 1.988-63.60 g/mL. The regression equation was: Y=10.765X+6.6433, r=0.9997. Two component precision tests RSD <1%; within 48 hours stability test RSD <1%; sample recovery test RSD <2%. Conclusion: This method is simple, rapid, accurate and reliable. It can simultaneously determine the contents of baicalin and glycyrrhizic acid in Xiaochaihu Decoction and provide reference for the effective control of Xiaochaihu Decoction.
