拟南芥AtFtsHi基因家族在叶绿体的发育中具有重要作用。它的成员AtFtsHi4 T-DNA突变体胚胎致死,这严重制约了对其生物学功能的研究。为了研究AtFtsHi4的功能,本文构建了AtFtsHi4的RNA干扰载体并通过转化拟南芥获得转基因植株。对转基因植株的表型进行分析,结果表明:与野生型相比,T3代RNAi转基因植株的子叶、叶片和果荚均出现了不同程度的黄化;叶绿素含量仅为野生型的一半;叶绿体的基粒垛叠数和基质片层数减少。这些结果表明,AtFtsHi4基因表达量的降低会抑制叶绿体类囊体的形成,进而影响叶绿体的发育。“,”AtFtsHi family plays an important role in chloroplast development in Arabidopsis thaliana. However, its member AtFtsHi4 T-DNA knockout line exhibits the embryonic lethality, which restricts the functional characte-ristics of this gene. In order to investigated the function of AtFtsHi4, in this paper we constructed AtFtsHi4 RNA interference vectorand produced its transgenic Arabidopsis thaliana. Phenotype analysis revealed that AtFtsHi4 RNAi transgenic plants had the etiolated cotyledons, leaves, siliques, one half of the chlorophyll content, and fewer grana stacks, stroma lamellae compared with the wild type. These results suggest that decreasing of AtFtsHi4 expression inhibits thylakoid membranes formation, thereby affects the chloroplast development.
