目的比较分析鼻咽癌患者与健康人群中血清中特征性蛋白质质点的差异,为鼻咽癌患者的诊断及侵袭转移预测提供参考及指导。方法选取2014年12月—2015年12月经本院收治的53例鼻咽癌患者以及同期体检的100名健康人群的血清标本作为研究对象,利用质点自动识别的蛋白质双向电泳(2 dimensional electrophoresis)和基质辅助激光解吸飞行时间质谱(MALDI-TOF-MS)技术分析鼻咽原发癌与淋巴结转移癌组织、鼻咽癌患者血清与正常人血清中特征性差异蛋白质质点差异。结果通过基质辅助激光解吸飞行时间质谱及酶联免疫吸附法(ELISA)验证得到sICAM-1、SAA、及HSP70等3种特征性差异蛋白且3种蛋白在鼻咽癌患者中的分布明显高于正常对照组中的分布,在鼻咽癌有转移组中的分布明显高于鼻咽癌无转移组中的分布,差异有统计学意义(P<0.05)。结论 sICAM-1、SAA、及HSP70等3种特征性差异蛋白可作为临床上针对鼻咽癌诊断及侵袭转移预测的蛋白指标,针对3种蛋白进行检验有利于临床上鼻咽癌患者诊断及个体化治疗的开展。
Objective To compare and analyze the differences of the characteristic protein spots in the serum of patients with nasopharyngeal carcinoma and healthy people and provide reference and guidance for the diagnosis, invasion and metastasis prediction of patients with nasopharyngeal carcinoma. Methods Serum samples from 53 patients with nasopharyngeal carcinoma and 100 healthy subjects who were admitted to our hospital from December 2014 to December 2015 were enrolled in this study. Two-dimensional electrophoresis (SDS-PAGE) Matrix-assisted laser desorption / emission-time-of-flight mass spectrometry (MALDI-TOF-MS) was used to analyze the differences of the characteristic protein spots in the serum of the patients with primary nasopharyngeal carcinoma, lymph node metastasis and serum of nasopharyngeal carcinoma patients. Results The three differentially expressed proteins, sICAM-1, SAA and HSP70, were identified by matrix-assisted laser desorption / ionization time of flight mass spectrometry and enzyme-linked immunosorbent assay (ELISA). The distribution of three proteins was significantly higher in patients with NPC The distribution in the normal control group was significantly higher than that in the non-metastatic NPC group (P <0.05). CONCLUSIONS: Three different characteristic proteins, sICAM-1, SAA and HSP70, can be used as clinical indicators for the diagnosis of nasopharyngeal carcinoma and the prediction of invasion and metastasis.
