目的 :研究 4 硝基喹啉氧化物 (4 nitroquinoline 1 oxide ,4NQO)诱发大鼠腭粘膜上皮细胞癌变过程中细胞内微量元素的变化及意义。方法 :将 4NQO涂于 2 8只Wistar大鼠硬腭后部粘膜 ,每周三次 ,共 19周。不同时期的所有粘膜标本被病理学分别证实为正常粘膜 ,癌前病变及鳞状上皮细胞癌 ,并进行电子探针显微分析。结果 :鳞状上皮细胞癌核及胞浆内铜 (Cu) ,锌 (Zn)明显下降 (P <0 .0 5 )。癌前病变上皮细胞浆内钼明显减少 (P <0 .0 1) ,而鳞癌细胞胞浆内Mo明显升高。癌变过程中 ,细胞内Se无明显变化。同时 ,鳞癌细胞Cu/Zn升高 ;Cu/Se ,Zn/Se降低。结论 :细胞内Cu ,Zn ,Mo的改变与口腔癌变有明显关联。
Objective: To study the changes of intracellular microelements during the carcinogenesis of rat palatal mucosa epithelial cells induced by 4-nitroquinoline 1 oxide (4NQO). METHODS: 4NQO was applied to the posterior mucosa of 28 Wistar rats three times a week for 19 weeks. All mucosa specimens from different periods were confirmed by pathology as normal mucosa, precancerous lesions and squamous cell carcinoma, respectively, and electron microscopy. Results: The squamous cell carcinoma nuclei and intracytoplasmic copper (Cu), zinc (Zn) decreased significantly (P <0.05). Premalignant lesions of epithelial cells were significantly reduced molybdenum (P <0.01), while the squamous cell cytosolic Mo was significantly higher. In the process of carcinogenesis, intracellular Se did not change significantly. At the same time, Cu / Zn in squamous cell carcinoma increased; Cu / Se, Zn / Se decreased. Conclusion: The changes of intracellular Cu, Zn and Mo are obviously associated with oral carcinogenesis.
