BT癌系是用苯并芘在移植到裸鼠皮下的人胎儿支气管内诱发的肿瘤。为了深入了解苯并芘的致癌机理,我们对该癌系中p ̄(53)基因的改变进行了系统的研究。结果表明:BT细胞核内p ̄(53)蛋白异常高表达;PCR-SSCP检测到第7外显子有异常改变;DNA序列分析证实第248密码子发生了CGG→CTT的颠换,所编码的氨基酸由精氨酸变为亮氨酸。本实验提示该突变可能是苯并芘引起癌变的重要分子基础。
The BT cancer line is a bronchus-induced tumor of a human fetus transplanted subcutaneously in nude mice with benzopyrene. In order to gain insight into the carcinogenic mechanism of benzo(a)pyrene, we systematically studied the alteration of the p53 gene in this cancer line. The results showed that the p ̄ (53) protein in BT cell nuclei was abnormally high; PCR-SSCP detected abnormal changes in exon 7; DNA sequence analysis confirmed that translocation of CGG → CTT occurred in the 248th codon. The amino acid is changed from arginine to leucine. This experiment suggests that this mutation may be an important molecular basis for benzopyrene-induced canceration.
