【目的】探讨中药复方开心胶囊对大鼠心梗后心室重构的影响及作用机理。【方法】结扎左冠状动脉主干造成大鼠心梗后心室重构模型 ,术后 3d给予开心胶囊 (西洋参、黄芪、麦冬、川芎、香附等组成 ) 8周后 ,用羟脯氨酸法及放免法检测大鼠非梗塞区左室心肌胶原总量及循环和心肌局部血管紧张素Ⅱ、醛固酮含量。【结果】模型组大鼠左室非梗塞区胶原总量高于假手术组 (P <0 0 1)。开心胶囊高、低剂量组胶原总量较模型组降低 (P <0 0 1) ,与开搏通组相近 (P >0 0 5 )。【结论】开心胶囊可以抑制胶原蛋白的合成 ,增加心肌顺应性。对肾素—血管紧张素—醛固酮系统的影响可能是其作用机制之一。
【Objective】 To explore the effect and mechanism of traditional Chinese medicine compound Kaixin capsule on ventricular remodeling after myocardial infarction in rats. 【Methods】 The ventricular remodeling model was established after ligation of the left main coronary artery in the left ventricle of rats. After 3 weeks, Kaixin capsule (American ginseng, Astragalus membranaceus, Ophiopogon japonicus, Chuanxiong, Xiangfu, etc.) was given and the hydroxyproline method was used. The total amount of collagen in the left ventricular myocardium and the contents of local angiotensin II and aldosterone in the left ventricular myocardium were measured by radioimmunoassay. 【Results】 The total amount of collagen in the left ventricular non-infarcted area was higher in the model group than in the sham group (P <0 01). The total amount of collagen in the high- and low-dose groups of Kaixin capsule was lower than that in the model group (P <0 01) and similar to that in the captopril group (P > 0 05). 【Conclusion】 Kaixin capsule can inhibit collagen synthesis and increase myocardial compliance. The effect of renin-angiotensin-aldosterone system may be one of its mechanisms.
