关于水稻与近缘稻种关系的研究方法,生物学上已有多种学说,由于目前国内外的近缘水稻rDNA的(internal transcribed spacers,ITS)以及他们的二级结构并未进行很多研究,所以本研究拟选取ITS序列作为一个分子分析指标,对他们的亲缘关系进行探索。我们用PCR扩增的方法获得了ITS,并进行PCR产物的克隆测序材料选取包括广陆矮四号稻、药用野生稻、宽叶野生稻、高杆野生稻四种。对以上四种稻的rDNA内ITS(ITS1+ITS2)以及5.8s rDNA序列进行测定和分析。最后,本文还用软件对栽培稻与这几种野生稻ITS2的二级结构进行了预测。
There are many theories about the research methods of the relationship between rice and its close relatives. Since many researches have not been done on the internal transcribed spacers (rDNA) and their secondary structure at home and abroad, Therefore, this study intends to select ITS sequence as a molecular analysis of indicators to explore their genetic relationship. We obtained the ITS by PCR amplification and cloned the PCR products. The selected materials include four varieties of rice, medicinal wild rice, medicinal wild rice, broad-leaved wild rice and wild rice of Gaogeng. The rDNA ITS (ITS1 + ITS2) and 5.8s rDNA sequences of the above four rice species were determined and analyzed. Finally, the paper also used software to predict the secondary structure of cultivated rice and ITS2 of these wild species.
