目的:探讨荔枝核含药血清对人肝癌HepG2细胞增殖和凋亡的影响。方法:采用血清药理学方法制备荔枝核、环磷酰胺含药血清以及对照血清,作用于HepG2细胞,MTT法检测各种血清对HepG2细胞增殖的影响,Hochest33258荧光染色观察细胞凋亡,流式细胞仪(FCM)检测细胞凋亡率。结果:荔枝核含药血清处理HepG2细胞后,呈剂量依赖性抑制肿瘤细胞增殖活性;并出现细胞核浓染与核碎裂;流式细胞仪检测HepG2细胞凋亡率明显高于对照组。结论:荔枝核含药血清能抑制人肝癌HepG2细胞增殖,其机制可能与诱导HepG2细胞凋亡有关。
Objective: To investigate the effect of Litchi seed serum on the proliferation and apoptosis of human hepatoma HepG2 cells. METHODS: Serum and pharmacological methods were used to prepare litchi nucleus, cyclophosphamide-containing serum and control serum for HepG2 cells. MTT assay was used to detect the effect of various serum on the proliferation of HepG2 cells. Hochest33258 fluorescence staining was used to observe apoptosis and flow cytometry. Instrument (FCM) detects the rate of apoptosis. RESULTS: After treatment with Litchi seed serum, HepG2 cells inhibited the proliferation of tumor cells in a dose-dependent manner. Nuclei staining and nuclear fragmentation appeared. The apoptosis rate of HepG2 cells detected by flow cytometry was significantly higher than that of the control group. CONCLUSION: Litchi seed serum can inhibit the proliferation of HepG2 cells, which may be related to the induction of HepG2 cell apoptosis.
