建立山慈菇药材中dactylorhin A和militarine的RP-HPLC含量测定方法,为山慈菇药材的质量标准研究提供依据。采用Alltech Prevail C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈(A)-水(B)为流动相进行梯度洗脱(0 min,20∶80;30min,55∶45;35 min,55∶45)。流速为0.8 mL·min-1;检测波长为224 nm;柱温为30℃。dactylorhin A和militarine的线性范围分别为0.257~9.95μg(r=0.999 8),0.128~10.27μg(r=0.999 9);平均回收率(n=9)分别为94.70%,102.8%,RSD分别为4.2%,4.9%。采用RP-HPLC测定来源于独蒜兰和云南独蒜兰的山慈菇药材中dactylorhin A和militarine的含量,该方法准确、重复性好,适用于来源于独蒜兰和云南独蒜兰的山慈菇药材的质量控制。
RP-HPLC method was established for the determination of dactylorhin A and militarine in Agrocybe punctatus, which provided the basis for the quality standard research of Agrocybe. The chromatographic column was eluted with Alltech Prevail C18 (4.6 mm × 250 mm, 5 μm) with gradient elution with acetonitrile (A) -water (B) (0 min, 20:80, 30 min, 55:45, 35 min, 55:45). The flow rate was 0.8 mL · min-1; the detection wavelength was 224 nm; the column temperature was 30 ℃. The linear range of dactylorhin A and militarine were 0.257-9.95μg (r = 0.999 8) and 0.128-10.27μg (r = 0.999 9) respectively. The average recoveries (n = 9) were 94.70% and 102.8% 4.2%, 4.9%. The content of dactylorhin A and militarine in Shanzijiao from Medicinal plants of Rhizoma et Radix and Yunnan yunnan was determined by RP-HPLC. The method is accurate and reproducible. It is suitable for the determination of dactylorhin A and militarine derived from Rhizoma Dioscoreae Quality control of Sagittaria medicinal herbs.
